N-Demethyl rifampin is a prodrug form of rifampin that is metabolized by the liver to its active form. It has been shown to be effective against murine leukemia viruses, viroplasms and resistant strains of virus. N-Demethyl rifampin inhibits viral polymerase activity by binding to the enzyme's active site, which prevents it from forming an enzyme-substrate complex with the viral DNA template. This prevents RNA synthesis and DNA replication, inhibiting the production of new viruses. N-Demethyl rifampin also has an inhibitory effect on cellular proliferation in tissue culture and may be used as a cancer chemotherapeutic drug.
