Mifobate is a fatty acid that is structurally similar to prostaglandin E2. It has been shown to inhibit the activation of caspase-1 and increase mitochondrial membrane potential in cells with oxidative injury. It also reduces insulin sensitivity in wild-type mice but not in transgenic animals. Mifobate can be used as a pharmaceutical dosage for the treatment of obesity, type 2 diabetes, and metabolic syndrome. Mifobate binds to Toll-like receptor 4 (TLR4) on human monocytes and inhibits peroxisome proliferator-activated receptor gamma coactivator 1 alpha (PGC1α) activity. This inhibition leads to reduced insulin sensitivity and increased fatty acid release from adipose tissue.
