Gliotoxin is a naturally occurring, fungal metabolite that is structurally related to the antibiotic amphotericin B. Gliotoxin has been shown to inhibit mitochondrial membrane potential and induce toxicity in cultured cells. It is also toxic to isolated mitochondria and in mice infected with opportunistic fungal species. In vitro studies have shown that gliotoxin inhibits the growth of many different types of fungi, including Candida albicans, Aspergillus fumigatus, and Cryptococcus neoformans. Gliotoxin also has been shown to promote the expression of genes that are involved in cellular responses to stress, such as the production of heat shock proteins. These properties make it a promising lead compound for pharmacological agents against infectious diseases.
