GS-6620 is a non-peptide inhibitor of the NS3 protease. It is designed to be an analog of sofosbuvir, which is an inhibitor of the NS5B RNA polymerase and a prodrug for the treatment of chronic hepatitis C virus (HCV). GS-6620 has been shown to inhibit HCV in vitro and in vivo. The mechanism by which GS-6620 inhibits HCV is not yet known, but it may be due to its ability to bind to the NS3 protease and block its activity. GS-6620 has also shown anti-viral properties against other viruses such as HIV, influenza A/B, herpes simplex type 1 and 2, adenovirus type 5, vesicular stomatitis virus and rhinovirus.
GS-6620's inhibition of protein synthesis may be due to its interaction with toll-like receptor (TLR) 4 or TLR7 on cells
