Presented as a purified antigen in proprietary buffer, pH 10. This recombinant SARs-CoV-2 Spike S1-S2 chimeric protein has been expressed in insect cells and purified by chromatography. It is important to note that repeated freezing and thawing should be avoided and it is suitable for ELISA assays and similar solid phase formats.
The severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), causes the Coronavirus Disease-2019 (COVID-19) and has dominated the world’s scientific attention since it was first reported in December 2019. Symptoms, which may include shortness of breath and fever, result from SARs-CoV-2 binding to the host cell receptors: angiotensin-converting enzyme 2 (ACE2) present in the arterial smooth muscle cells and endothelial cells and additionally venous endothelial cells in organs. This interaction is carried out by the SARs-CoV-2’s spike protein. Of the two subunits that this spike protein is composed of, S1 binds the ACE2 receptors through its receptor binding domain while S2 initiates membrane fusion between the virus and host through forming a hairpin structure. While the spike protein subunits can be collectively used as the immunogen in vaccine development, the receptor binding domain in the S1 subunit is of particular interest for the development of neutralizing antibodies. Interestingly a proline substitution within the S2 domain enhances the immunogenicity of this spike protein.
