Description:The rubella virus is an airborne pathogen made up of a nucleocapsid core surrounded by E1 and E2 type 1 membrane glycoproteins forming heterodimer spikes through disulphide bonds. It is thought that the rubella virus' entry into human host cells endocytically, is dependent upon these ectodomain proteins E1-E2. A 28 residue hydrophobic domain within E1 enables E1-cell membrane fusion, while E2's positively charged sequence: RRACRR interacts with negatively charged phospholipids on the host lipid bilayer. In all Rubella strains both proteins have a transmembrane domain and E1 proteins have three N-linked glycosylation sites. However only M33 and HPV-77 strains have four N-linked glycosylation sites located within E2. Glycosylation of E1 proteins ensures its correct folding and ensures antigenic and immunogenic epitopes are expressed. Alternatively within E2 these N-linked sites are important in glycan processing and its overall stability. Individually E2 contains O-linked sugars and has hemagglutination and neutralizing epitopes while E1 has the ability to bind to myelin oligodendrocyte glycoprotein (MOG), a common central nervous system protein.
Upon entry into the host cell the low pH of the endosome vacuole conformationally changes E1 and E2, mediating their fusion to the membrane. Although the pathogen inflicts mild rash or fever symptoms, during pregnancy it may result in birth defects.This recombinant rubella glycoprotein ectodomain (E1-E2) of the HPV-77 strain expressed in insect cells, has been chromatographically purified and presented in a proprietary buffer formulation. In addition to its therapeutic application this ectodomain (E1-E2) is suitable for use in: ELISAs, bead based assays and similar solid phase formats.
