STO-609 is an orally bioavailable small molecule that inhibits the activity of a key enzyme in the insulin signaling pathway, protein kinase B (PKB). It has been shown to inhibit the growth of new blood vessels and reduce neuronal death. The drug was evaluated in vitro as well as in vivo for potential therapeutic effects on diabetes. In vitro, STO-609 inhibited PKB and suppressed glucose uptake. In vivo, STO-609 significantly lowered blood glucose levels and increased insulin sensitivity in mice with type 2 diabetes. The drug also showed inhibitory properties against a response element that is involved in the angiogenic process. These results suggest that STO-609 may be useful for treating diabetic patients who have not responded to other therapies.
