Biotinylated aMptD, a Mycobacterium avium subsp. Paratuberculosis (MAP) specific ligand. MAP can cause Johne disease (the wasting disease) in livestock. It is important therefore to detect the presence of MAP in animal milk and faeces.Biotinylated aMptD can be used (along with biotinylated aMp3) to detect the viability of MAP cells in infected livestock through combined peptide-mediated magnetic separation phage display due to their high affinity for MAP. The addition of biotin to aMptD glycine residue, changes the orientation of aMptD so that it can bind to the target bacteria with increased stability, thus achieving a high capture efficiency. A similar effect is observed on the addition of biotin to aMp3 asparagine residue.
