In its intact state this peptide is not fluorescent, however when this substrate peptide is cleaved, Rhodamine 110 is released and thus fluorescence can then be detected. The terminal carboxylic acids in this peptide have been activated by the addition of the p-tosyl moiety, allowing for easy addition of functional groups or further peptide residues.
