A Wnt agonist is a drug that can stimulate the activity of Wnt proteins. It is usually used to treat cancer. The Wnt protein family consists of 19 members, which are involved in many aspects of cellular physiology including cell specific functions and development. The most common use for Wnt agonists is to treat colorectal adenocarcinoma, which has been shown to be effective through the inhibition of tumor growth and metastasis.
Wnt agonists bind to the Frizzled receptors on the surface of cells and activate intracellular signaling pathways that result in changes in gene expression or other cellular processes. One way they do this is by inhibiting the production of proteins that induce apoptosis (cell death), such as caspase-independent cell death and autophagy, which may lead to tumor suppression. Another mechanism involves activation of Rho-associated kinase (ROCK) in response to WNT stimulation, leading to activation of downstream effectors
