MMP-3 is a protein that is active in the human body and plays an important role in the breakdown of extracellular matrix. It has been shown to be involved in the disease activity of multiple sclerosis and rheumatoid arthritis. MMP-3 is synthesized as a proenzyme, which contains a signal peptide that directs it to the endoplasmic reticulum (ER). The proenzyme can be activated by two mechanisms: autocatalytic cleavage or by proteolytic cleavage by other enzymes such as serine proteases or metalloproteinases. Calcium pantothenate inhibits MMP-3 activation by autocatalytic cleavage, while nonsteroidal anti-inflammatory drugs inhibit proteolytic cleavage.
