Naphthol AS nonanoate is a substrate for plasma esterases, and is hydrolyzed to naphthol AS by these enzymes. It has been detected in erythrocytes and leukocytes, but not in other tissues. Naphthol AS nonanoate stains the sporopollenin in pollen grains and can be used as a cytochemical technique for identifying pollen grains. The specificity of this technique is increased by using an esterase modifier such as acetic acid or sodium acetate. The enzyme-substrate reaction produces a pink color that can be detected with a microscope.
