AOP is a phosphonium salt and a HOAt derivative employed as a coupling reagent for peptide synthesis. AOP can be used in solution and solid-phase synthesis, is highly efficient with both linear and cyclic peptides, allows to achieve good yields, is a racemisation suppressant and has good reaction rates. AOP is soluble in DMF and requires presence of a base such as Hünig’s base. The drawback of using AOP is the formation of the toxic and carcinogenic by-product HMPA, therefore, it must be handled with care. The pyrrolidino derivative PyAOP is a good alternative to AOP as it achieves comparable performances, but generates a less toxic by-product.
