Arotinolol is a beta-blocker that has been shown to decrease blood pressure by blocking the effects of catecholamines at sympathetic nerve endings. It has been shown to be effective in reducing systolic blood pressure in patients with hypertension who have not responded to other therapies. Arotinolol is also used as an antihypertensive agent for the treatment of congestive heart failure and diastolic hypertension. It was approved by the FDA in 1981. Arotinolol is metabolized by hydrolysis to propranolol and water, which are excreted through urine. The drug binds to beta-adrenergic receptors and inhibits the action of catecholamines on these receptors, thereby preventing activation of adenyl cyclase and subsequent formation of cyclic AMP (cAMP). Arotinolol can also bind to calcium channels, which may account for its ability to lower blood pressure.
