- Splitomycin
Splitomicin is a small-molecule inhibitor that binds to the protein SIRT1 and inhibits its activity. It has been shown to have therapeutic potential in autoimmune diseases, such as type 1 diabetes, by preventing the production of inflammatory molecules. This drug also has cardiac effects, which include inhibition of the transcriptional regulator Nrf2 and erythrocyte membrane fluidity. The molecular docking analysis of this compound with SIRT1 suggests that it binds in a region where it can inhibit the enzyme's activity by blocking access to the active site. This binding is thought to be due to the presence of nitrogen atoms that form hydrogen bonds with the enzyme's active site residues. The redox potential of splitomicin is -0.12 V vs NHE at pH 7.4, suggesting that it may be able to reduce reactive oxygen species (ROS) in cells.
