PyBroP is a phosphonium salt used in peptide coupling chemistry, analogous to PyCloP. PyBroP shows high efficiency in the coupling of N-methylated amino acids and α,α-disubstituted amino acids, with excellent yields, no epimerisation and fast reaction rates. PyBroP is a stable solid which can be used in a one-pot procedure and is soluble in the solvents commonly used for peptide coupling chemistry. PyBroP is a derivative of the phosphonium coupling reagent BOP but contains a pyrrolidine moiety in place of the dimethylamine of BOP. The aim behind the design of PyBroP was to prevent the formation of the extremely toxic and carcinogenic by-product HMPA generated with BOP, but with comparable performances.
