Baclofen is a centrally acting muscle relaxant and anxiolytic drug that is used in the treatment of spasticity and other symptoms related to multiple sclerosis. It is also used in the treatment of chronic pain, such as low back pain, and alcoholism. Baclofen has been shown to be effective against chronic pain by inhibiting voltage-dependent calcium channels. This inhibition leads to an increase in GABA (gamma-aminobutyric acid) transmission at synapses, which decreases neuronal excitability and prevents synaptic dysfunction. Baclofen has been shown to have carcinogenic potential when administered chronically orally and should not be used for more than 3 months without medical supervision.
