Captafol is a sesquiterpene lactone that is used as an antimicrobial agent against bacteria, fungi, and protozoa. It is a potent inhibitor of bacterial DNA gyrase and topoisomerase I. Captafol also inhibits the synthesis of mitochondrial cytochrome c oxidase and ribosomes in mammalian cells. Captafol is not effective against bacteria with reduced susceptibility to fluoroquinolones such as Escherichia coli and Salmonella enterica serovar Typhimurium. This drug has been shown to be toxic to rat proximal tubule cells in toxicity studies. Captafol can be detected by fluorescence detection after sample preparation. The matrix effect was observed when red blood cells were incubated with captafol for 3 hours before analysis using flow cytometry. The cells were resistant to captafol-induced lysis if they were pretreated with hydrochloric acid or sodium chloride.
