Cyclosporin A (CSA) is a natural compound that is found in the cell membrane. It binds to a response element on the mitochondrial membrane and inhibits p-gp, leading to an increase in the mitochondrial membrane potential. CSA also has genotoxic effects and can cause oxidative injury by inducing detoxification enzymes. This compound has been shown to be effective against autoimmune diseases, such as multiple sclerosis. Cyclosporin A-d13 (CSA-d13) is a metabolite of CSA that can be used for research purposes because it contains a 13C label, which makes it easier to detect using analytical methods. CSA-d13 binds to p-gp and inhibits its activity through competitive inhibition. The chemical structure of this drug is similar to other substrates of p-gp, so it may have some inhibitory effects on this transporter.
