Perindoprilat is a prodrug that is metabolized in vivo to enalaprilat, its active form. It has been shown to inhibit the conversion of angiotensin I to angiotensin II and to block the binding of angiotensin II to AT1 receptors. This drug is used for the treatment of hypertension and congestive heart failure. Perindoprilat also inhibits cyclase activity at low concentrations, which may be due to its ability to bind competitively to the active site on these enzymes. In addition, perindoprilat has been shown to increase renal blood flow by increasing the production of nitric oxide and prostaglandins. The effects on blood pressure are linear over a wide range with a calibration curve that can be used for monitoring purposes.
