PEG4-SPDP is a crosslinker used to attach proteins, siRNA, and other molecules to surfaces or other proteins. It consists of a polyethylene glycol (PEG) spacer arm and a disulfide bond that can be cleaved by reducing agents. PEG4-SPDP has been used to conjugate transferrin to the surface of Pseudomonas aeruginosa, allowing for targeted delivery of drugs or other payloads to cells expressing transferrin receptors. Additionally, it has been utilized in breast cancer research as a ligand for targeting cancer cells specifically while leaving healthy cells unaffected. The specificity of this targeting has been demonstrated through experiments in which PEG4-SPDP was used to crosslink transferrin onto the surface of breast cancer cells, resulting in increased uptake of drugs or other molecules by these cells compared to healthy cells.
