DCC is a carbodiimide coupling reagent for amide and ester formation, used in peptide synthesis in presence of benzotriazole-based additives such as HOBt or HOAt to limit epimerisation. The urea by-product is highly insoluble in water and most organic solvents; therefore it can be easily removed by filtration. The insolubility of the urea makes the use of DCC unsuitable for solid-phase peptide synthesis unlike DIC, and only suitable for solution-phase couplings. Carbodiimide reagents including DCC tend to generate an N-acylurea by-product, whose formation can be controlled by forming the activated ester first and adding the amine afterwards, together with low reaction temperatures and the choice of DCM or chloroform as solvent.
