7,9-Dimethylguanine is a modified nucleobase that is used in the synthesis of oligonucleotides. It has been shown to stabilize the DNA duplex and reduce the formation of undesired products. 7,9-Dimethylguanine is also able to form hydrogen bonds with other nucleobases, which may be due to its high proton affinity. This property allows it to function as an analog for guanine in DNA oligonucleotides. 7,9-Dimethylguanine can also form hydrogen bonds with anions such as chloride or bromide. The X-ray crystal structures of 7,9-dimethylguanine have been determined and show that it has a molecular conformation similar to that of guanine, but with a methyl group at position 9.
