2-(Dimethylamino)nicotinonitrile is a precursor for the synthesis of aminopyridines. It is prepared by diazotizing 2-amino-5-nitropyridine with sodium nitrite in concentrated hydrochloric acid, followed by addition of dimethylformamide and trifluoromethanesulfonic acid. The reaction yields 2-(dimethylamino)nicotinonitrile, as well as other products including 2-methylpiperidine and 4-methylpiperidine. This process can be improved by using microwaves or a catalyst such as trifluoromethanesulfonic anhydride or trifluoroacetic anhydride.
2-(Dimethylamino)nicotinonitrile is used to synthesize aminopyridines, which are intermediates in the production of various medications such as antibiotics and cancer drugs.
