Carbon disulphide is used in this reaction as it can react with phosphorus to form a reactive intermediate, which reacts with the phosphate group on the enzyme. This reaction is shown below:

Phosphonium ion + Active Site → Phosphineoxide radical + Inactive Site
The phosphineoxide radical attacks the phosphate group on the enzyme, leading to its inactivation. The high values for dichlorophenylphosphine are due to its high binding constants and low expression levels.
