2,2-Difluoro-2-chloroacetophenone (DF2CP) is a nucleophilic imidazole derivative that is synthesized from trifluoromethylated aniline. DF2CP reacts with cholinesterases to form reactive intermediates that bind to the active site of the enzyme and inhibit acetylcholine esterase, resulting in increased acetylcholine levels. It has been shown to be effective in animal models of Alzheimer's disease and Parkinson's disease. DF2CP reversibly binds to sulfonic acid groups on the enzyme surface and forms a hydrogen bond with the amine group in substrate binding sites. The substitution of fluorine for chlorine at C1 or C3 results in decreased solubility, while substitution at C5 increases solubility due to increased electron density.
