3,5-Dimethyl-4-nitroisoxazole is a protonated form of an activated methylene group. The protonated form has been observed in simulations of constant pressure and temperature conditions. This protonated form is the most reactive and has anticancer activity. It binds to DNA by alkylation and prevents RNA synthesis. 3,5-Dimethyl-4-nitroisoxazole is also synthesized by nitroaldol reaction with sodium methoxide in methanol or acetone at room temperature for 30 minutes.
