7,8-Dihydroxy-4-methylcoumarin is a natural compound that has been shown to have pharmacological activity. It is a potent inhibitor of cyclic nucleotide phosphodiesterases and has been shown to inhibit the proliferation of carcinoma cells in cell culture. 7,8-Dihydroxy-4-methylcoumarin inhibits the production of cyclic GMP (cGMP) in cells by competing with guanosine triphosphate (GTP) for binding to the catalytic site on phosphodiesterase type I. This inhibition results in an increase in cAMP levels in cells and an increase in mitochondrial membrane potential. 7,8-Dihydroxy-4-methylcoumarin also inhibits the synthesis of human immunoglobulin by inhibiting DNA polymerase.
