Endomorphin-2 is a cyclic peptide that is the endogenous ligand for the neurokinin-1 receptor and kappa-opioid receptors. It has been shown to have analgesic, anti-inflammatory, and antidiarrheal properties. Endomorphin-2 also has been shown to inhibit platelet aggregation and vasoconstriction in vivo. The biological activities of endomorphin-2 are similar to those of endomorphin-1, but it differs structurally in that it contains a single intramolecular hydrogen bond between the amide nitrogen of Tyr and Pro. This intramolecular hydrogen bond may be responsible for the high potency of endomorphin-2, as well as its conformational stability.
