Description:Ethyl 3-chloromethylbenzoate is an ester prodrug of acyclovir that is hydrolyzed to the active form in human plasma. It is a prodrug, which means that it is not the biologically active form of acyclovir. The ester bond in ethyl 3-chloromethylbenzoate is cleaved by hydrolysis, releasing the carboxylic acid and ethyl alcohol. The carboxylic acid is then converted to acyclovir by oxidation. Acetylation of the 3-position on the benzene ring increases its stability and enhances water solubility, making it more suitable for topical administration. The half life of ethyl 3-chloromethylbenzoate in humans has been shown to be 5 hours, which makes it more suitable for once daily dosing as opposed to other prodrugs with half lives as short as 1 hour.
