Description:

Fluorescein is a fluorescent molecule that belongs to the group of fluorescence probes. It is used in biology as a fluorogenic substrate for intracellular pH measurements, and it can be used to detect the presence of calcium ions. Fluorescein reacts with p-hydroxybenzoic acid and benzalkonium chloride to form fluorescamine, which has the ability to react with nucleophilic groups such as amines, thiols, and hydroxyl groups. The reaction mechanism of fluorescein with these groups is not well understood. It is hypothesized that it may involve an electron transfer from the aromatic ring of fluorescein to the nucleophile. This reaction produces reactive radicals that are capable of reacting with other molecules via radical addition or hydrogen abstraction reactions.
