When guanine HCl is protonated, it has a distinctive fluorescence spectrum with peaks at around 390 nm and 410 nm that can be measured using fluorescence spectrometry. The major metabolite of guanine HCl is thioguanine (TG), which can be detected using monoclonal antibody tests or by measuring malonic acid levels in serum samples.
