• Description: Ganaxolone is a therapeutic agent that is used to treat epilepsy. It binds to the GABA receptor and blocks the action of GABA, which is an inhibitory neurotransmitter. This binding prevents GABA from binding to the receptor and reduces neuronal excitability in the brain. The pharmacological effects of ganaxolone are irreversible because they are due to the formation of covalent bonds with basic proteins in neurons. Ganaxolone has been shown to increase locomotor activity in rats, but not affect performance on a rotarod test or basic sensory and motor abilities. The effect of ganaxolone on human macrophages has been demonstrated using an analytical method that measures the release of reactive oxygen species (ROS) from these cells. The long-term efficacy of ganaxolone has been assessed by evaluating its ability to prevent seizures in patients with refractory epilepsy who have not responded adequately to other treatments.
