Gramicidin is a polymyxin antibiotic that is used in biological studies. It has shown to have antibacterial efficacy against Gram-positive bacteria and some Gram-negative bacteria. The membrane-disrupting and antimicrobial activity of gramicidin is due to its ability to form hydrogen bonding interactions with the phospholipids in the bacterial cell membrane and transfer reactions with the phosphate groups of ATP. This interaction leads to the disruption of the membrane, which disrupts cellular functions such as energy metabolism and protein synthesis. Gramicidin also binds to the enzyme ATPase, which regulates ATP levels in cells, leading to an increase in ATP hydrolysis.
