Minodronate is a bisphosphonate drug that inhibits bone resorption by inhibiting the enzyme farnesyl diphosphate synthase. It also has antioxidative properties and slows the progression of age-related osteoporosis. Minodronate monohydrate has been shown to inhibit the growth of subcutaneous tumors in mice. The structural analysis of minodronic acid, the active form of minodronate, shows that it binds to the active site of farnesyl diphosphate synthase and prevents the conversion of dimethylallyl pyrophosphate to isopentenyl pyrophosphate. This reaction is necessary for normal cell division and protein synthesis. The plate test showed that minodronates inhibited collagen production with an IC50 value of 3.2 μM, which was comparable to that of alendronate (IC50=1.5 μM).
