3-Hydroxy desloratadine is a metabolite of desloratadine. It is one of the metabolites that can be formed from the drug in vivo, after oral administration. 3-Hydroxy desloratadine has been shown to be an active metabolite in vitro and in vivo, with linear calibration curves (R2>0.99) for both human liver and human plasma samples. The metabolic profile of 3-hydroxy desloratadine is similar to that of desloratadine, including inhibition of cytochrome P450 enzymes and glucuronidation by glucuronidases. In vitro studies have shown that 3-hydroxy desloratadine is not a substrate for CYP2D6 or CYP3A4, but it may inhibit CYP1A2 and CYP2C19. It has also been shown to be a substrate for UGT1A6 and UGT2B7.
