Loratadine is a nonsteroidal antihistamine that competitively blocks histamine H1 receptors in the brain, which decreases the allergic response. It also has glucuronide metabolites, which are eliminated by the kidney and can be measured in the plasma concentration. Loratadine's time-dependent effect on histamine release is due to its ability to block histamine H1 receptors for up to 12 hours after administration. Loratadine has been shown to have an antagonistic effect on clopidogrel, meaning it prevents the drug from working properly by blocking its action at the H1 receptor. This interaction could result in increased risk of adverse events, such as heart attack or stroke.
