3-Hydroxyacetaminophen is a reactive metabolite of acetaminophen. It is produced by the bacterial strain Acinetobacter calcoaceticus, which was found to produce 3-hydroxyacetaminophen in the presence of oxygen and nitrate. The production process can be optimized by using surface methodology and kinetic studies. In vitro assays showed that 3-hydroxyacetaminophen has a higher cytotoxicity than acetaminophen, as well as an inhibitory effect on tyrosinase activity. This may be due to the deuterium isotope effect, which causes a substitution of hydrogen with deuterium atoms. The reaction intermediates are not known, but they are thought to be generated by chemical reactions involving the substrate or other reactants.
