Hematoxylin is a basic dye that binds to the amino acid histidine in proteins, and is used as a stain in histology. It has been shown to be effective in the diagnosis of squamous cell carcinoma. Hematoxylin reacts with basic proteins by forming a bond with the amino group on the protein molecule. This reaction occurs more quickly at acidic pH levels, which are found in areas of inflammation or cancer cells. The rate constant for this reaction is high, making it useful for identifying these cells. Benzalkonium chloride (BAC) is an antiseptic agent that can inhibit hemolysis and proteolysis of erythrocytes during staining procedures.
