1,2-Isonicotinylhydrazine (1,2-INH) is a mutagen that inhibits the polymerase chain reaction. It is an amide product of nicotinic acid and hydrazine. 1,2-INH has been shown to be mutagenic in vitro. It also exhibits antitubercular activity against Mycobacterium tuberculosis by inhibiting DNA synthesis. 1,2-INH binds to the RNA polymerase of M. tuberculosis and inhibits its acetylation activity. This leads to inhibition of transcription and replication of DNA. The compound also binds to the ligand binding site of the enzyme and causes changes in the magnetic properties of its metal ion cofactor, which may lead to structural changes in the polymerase complex.
