10-Ketonaltrexone is an opioid receptor antagonist that binds to the μ- and δ-opioid receptors in the gastrointestinal tract, blocking the effect of opioids. It has been shown to selectively inhibit binding of oxycodone, cyclazocine, and morphine to their receptors in muscle and deferens tissue. 10-Ketonaltrexone has affinity for opioid receptors in ileum tissue and binding sites in the brain. This drug is selective for the μ-receptors over δ-receptors and has no activity against other receptors such as dopamine, serotonin, or histamine. In vitro assays have shown that 10-ketonaltrexone binds with high affinity to human naltrexone binding sites but not to human oxymorphone binding sites.
