Taletrectinib is a potent and selective small molecule activator of the MET receptor tyrosine kinase. It has been shown to have anti-cancer activity in preclinical models of cancer by inducing tumor cell apoptosis, inhibiting tumor growth, and preventing metastasis. Taletrectinib has also been shown to inhibit the activation of downstream signaling pathways that are important for cancer cell proliferation, including PI3K/AKT and MAPK/ERK. Ligand binding experiments conducted on recombinant MET indicate that taletrectinib binds to an allosteric site on MET and inhibits receptor activation with an IC50 value of 0.5 nM.
