2-Methyl-3,5-dinitroaniline is a chemical that is used as an intermediate in the production of other chemicals. It has been shown to cause genotoxic effects at high doses in tissue culture and to inhibit the activity of certain enzymes by binding to sulfhydryl groups. 2-Methyl-3,5-dinitroaniline also has a dose dependent effect on the uptake of radioactive compounds into cells. This chemical inhibits protein synthesis by acting as a competitive inhibitor for ribonucleotide reductase (RNR) and DNA polymerase II, which are involved in the synthesis of nucleic acids. The inhibition of these two enzymes leads to cell death through apoptosis. The genotoxicity and inhibition of RNR and DNA polymerase II have been shown in both untreated control cells and V79 cells with monoclonal antibodies against these enzymes. The analytical method used for this analysis was HPLC with fluorescent detection.
