Methyl 4-bromothiophene-3-carboxylate (MBT) is a lactam compound that has shown antimalarial activity. It is an inhibitor of the enzyme dihydrofolate reductase, which is essential for malaria parasite survival. MBT has been shown to be active against both chloroquine-sensitive and -resistant strains of plasmodium falciparum, as well as against aminopyrimidines. MBT can be synthesized in two steps from commercially available starting materials. The first step involves reduction of 4-bromothiophene-3-carboxylic acid with lithium aluminum hydride to give methyl 4-bromoacetate, which undergoes a Suzuki coupling with methylamine to give the desired product. The second step entails cyclization of methyl 4-bromoacetate with potassium hydroxide to produce the desired product.
