(R)-Monophos is an antitumor agent that inhibits the growth of cancer cells. It is a potent inhibitor of DNA synthesis, which is essential for cell division. (R)-Monophos has been shown to be effective against a number of cancer cell lines, including leukemia, lymphoma and breast cancer cells. This drug is synthesized by the cross-coupling reaction between a phosphine and an activated carbonyl group. The mechanism of this reaction involves the formation of an iminium ion intermediate with phosphite as the leaving group. The iminium ion reacts with a chloroformate to produce (R)-monophos and hydrogen chloride as byproducts. Crystal x-ray diffraction data have provided evidence for this mechanism.
