3-O-Methyl estrone is a trifluoroacetic acid derivative of oestrone that is used in the synthesis of premarin. 3-O-Methyl estrone has chemical reactions that are similar to those of oestrone, but it also has a hydroxyl group on the C3 position. The product research and development of 3-O-methyl estrone led to discovering its structural similarity to estradiol and its potential as an estrogen receptor modulator. This compound showed high affinity for the estrogen receptor alpha (ERα) and ERβ, with binding constants of 1.9 nM and 0.87 nM, respectively. Molecular modeling studies revealed that 3-O-methyl estrone has a higher affinity for ERα than for ERβ. Kinetic data suggests that this compound could be potentially used in hormone replacement therapy or to treat osteoporosis in postmenopausal women.
