(2S,4S)-4-Methylglutamic acid is a synthetic glutamate analog that can bind to the active site of daptomycin. It has been shown to have a higher binding affinity than natural glutamate and is stable at high temperatures. This compound has been evaluated in binding experiments and functional assays against carbapenem-resistant Gram-positive bacteria, including methicillin resistant Staphylococcus aureus (MRSA) and Enterococcus faecium. The synthesis of this compound was achieved by solid-phase synthesis, which used commercially available amino acids. The chemical profile of this analog was also evaluated using various assays, such as uptake assays on retinal neurons. A synthetic scheme was developed for the preparation of this compound.
