Description:3-(Methylnitrosamino)propionitrile (NNK) is a potent, but toxic, carcinogen that has been shown to be one of the major causes of lung cancer in animals. It is metabolized to 3-aminobenzamide and N-nitrosodimethylamine (NDMA) by cytochrome P450 enzymes. These metabolites are then further converted into reactive species that bind covalently with DNA, leading to mutations and ultimately cancer. The activation of NNK can be prevented by inhibiting the enzyme hydrolysis or by blocking the synthesis of the reactive metabolites.
