Methyl-4-iodobenzoate is an organic compound that can be used as a precursor to the synthesis of pemetrexed, a drug that has been shown to be effective against cancer. Methyl-4-iodobenzoate is synthesized by combining 4-iodobenzoic acid with methyl iodide in the presence of sodium azide. The reaction mechanism is thought to involve the formation of an aziridine intermediate and proceeds via a suzuki coupling reaction. This compound can also be synthesized through hydrolysis of methyltriazole and azetidinium chloride with potassium hydroxide, followed by chlorination with hydrochloric acid. Methyl-4-iodobenzoate is soluble in organic solvents such as chloroform and ether, but not in water.
