Nitrofurantoin is an antibiotic that inhibits bacterial growth by inhibiting protein synthesis. It binds to the ribosomal 50S subunit, preventing the formation of an antibiotic-inhibitor complex with the enzyme cell wall synthesis that is required for cell wall biosynthesis, inhibiting protein synthesis and cell division. Nitrofurantoin has been shown to be effective against wild-type strains of bacteria, but not against drug-resistant strains. The rate constant for nitrofurantoin binding to ribosomal RNA has been determined using a polymerase chain reaction assay on tetracycline-resistant bacteria. A long-term toxicity study in rats has demonstrated that nitrofurantoin can cause toxic epidermal necrolysis and necrosis at high doses. Nitrofurantoin also has some biochemical properties that are similar to those of natural compounds such as colloidal gold and have been used as a model system for studying antibiotics' interactions with mammalian cells.
